Proteins, says Alex Rives, “are really the basis of our biology—the building blocks of life. Our genes encode proteins, and proteins build the complex dynamics of a cell. So if you think about life, life is fundamentally programable.” But understanding the language of that protein program—how they are built, the different shapes they can take, and the various functions that those structures then have in the human body—has remained a biological black box. 

As a computer scientist, Rives sees great potential in proteins: if programmed and then made more intelligible, perhaps they could create new ways of understanding and treating disease. “Proteins have really deep patterns in both their sequence and structure, so I thought that there might be something about proteins that might be learnable,” he says. 

At Meta, Rives pioneered Evolutionary Scale Modeling (ESM), the first language model for understanding proteins that represented a breakthrough in bridging computer science and biology. Rives is now head of science at the [Chan Zuckerberg Initiative](https://time.com/4503222/mark-zuckerberg-priscilla-chan-initiative-cure-diseases/), a philanthropic health initiative with the goal of using technology to cure all disease by the end of the century. Earlier this year, he revealed the latest iteration of ESM: ESMFold2, a database of a billion proteins and their structures. Understanding protein structure is key to understanding their function, and the database is already a powerful tool for drug developers who depend on protein structure to predict which could treat and even cure disease. 

By shifting more of drug development from manually tedious, lab-intensive processes to models generated via digital representations and predictions, coming up with new drugs could become not just more efficient, but more bespoke. “There are certain areas like rare diseases and cancers where the disease really differs substantially among individuals,” Rives says. “There is a real potential there in being able to have therapies we can really program for every single patient in silico.”
